Objectives:The primary objective of this study was to investigate the pharmacokinetics and pharmacodynamics of Orlistat (Xenical®) in Chinese patients with obesity and associated diabetes mellitus. The secondary objectives were to determine the pharmacokinetics and pharmacodynamics of Orlistat in Chinese patients with obesity and related diabetes mellitus and related metabolic diseases. The study design was a single-center, double-blind, placebo-controlled, placebo-controlled study. Patients with obesity or diabetes mellitus and associated metabolic diseases and those with diabetes mellitus, or those with obesity and associated diabetes mellitus, or those with diabetes mellitus, or other causes of obesity and diabetes mellitus and related metabolic diseases were included in the study. Orlistat is an oral prescription drug approved by the U. S. Food and Drug Administration (FDA). It is available as a single-dose oral capsule or as an intramuscular injection and is used to treat approximately 30 million patients in the U. alone. Orlistat is rapidly absorbed and eliminated rapidly in the gastrointestinal tract, and peak plasma levels are reached in approximately 2-4 hours. The half-life of Orlistat is 6.8 hours. The pharmacokinetic parameters of Orlistat were evaluated in a single-center, double-blind, placebo-controlled, crossover, randomized, crossover, study in which patients were divided into three groups: a placebo group, a Group A (oral capsule or intramuscular injection), and a Group B (oral capsule or injection) group. Orlistat (Xenical®) is a lipase inhibitor. Orlistat is metabolized predominantly by the hepatic enzyme CYP2C19 and/or CYP3A4, with the predominant metabolite being orlistat. Orlistat is extensively metabolized by cytochrome P450 3A4, CYP3A, and CYP2D6 to andlistat and is partially hydrolyzed by CYP2C19 and CYP3A4, and in the presence of CYP2C19 and CYP2C19 and/or CYP3A4, is principally hydrolyzed by CYP2C19 and CYP2C19.
